Family-based Quasi-Likelihood Score test (FQLS)
FQLS is a powerful and efficient statistic for genetic association analysis that considered the heterogeneity of effect by ascertainment bias among family members, under the assumption that both prevalence and heritability of disease are available.
We have assumed that there was a well-designed set of individuals who were“probands”, and for this situation, we calculated the statistic as indicated and denoted FQLS1. In practice, the set ofprobandswill not be welldefinedunless the family contains only one affected member. For this situation, we calculated the probability for each individual to be affected under the assumption that all the other family members were“probands”, and the statistic calculated this way was denoted by FQLS2.
NOTE : FQLS only can be applied to dichotomous phenotype !
Multiple Family-based Quasi-Likelihood Score test (MFQLS)
MFQLS is a powerful and efficient statistic for genetic association analysis that considered correlated variables. It can be applied to the dataset having multiple phenotypes or muliple markers, such as gene set in population and family-based cohort. And it can be used both contious and binary phenotypes!
